The authors state 'When a strain expressing Cas9 and a TRI5 targeting guide RNA (DTRI465Cas9-sgRNA::TRI5) was used in place of the Spok1 insert in the TRI465 locus (Gardiner & Kazan, 2018) equal segregation (1 : 1, n = 16) was observed, suggesting strongly that this particular nuclease-based design, similar to high-efficiency drive systems shown to work in a number of other species including yeast and insects (DiCarlo et al., 2015; Hammond et al., 2016), is unable to drive in F. graminearum.'
This design is therefore designated as non-functional.
